How can I compute the latency in this simulator since I hope to simulate how long it will takes for an application to request a data packet and receive a data packet.

Hi,
for nr 5G-Lena module, you can use built-in scenario Flow Monitor (at the bottom of scenario).

For mmwave module, there is trace to file or E2 latency on PDCP layer.

For your specific request, packet tracing, i think you should build your own trace to file function in proper layer or at packet app side.
